Resumo |
As Novas Substâncias Psicoativas (NSPs) conhecidas como NBOHs são fenetilaminas substituídas e têm uma estrutura similar às NBOMes, o que torna desafiador o desenvolvimento de testes de detecção rápidos e seletivos para identificação inequívoca. Os testes colorimétricos são vantajosos na detecção de substâncias devido ao seu baixo custo, resultados rápidos, uso de reagentes estáveis e possibilidade de aplicação in loco. Além disso, devido à sua recente inserção no mercado ilegal para uso abusivo, são poucos os estudos que descrevem os efeitos potencialmente tóxicos decorrente do uso dessas substâncias. Portanto, o presente estudo apresenta dois objetivos: 1) desenvolver um método colorimétrico de aplicação em campo para a detecção seletiva de NBOHs em drogas apreendidas e 2) realizar estudos iniciais sobre a toxicidade da substância chamada MD-NBOH. Inicialmente, os analitos 25H-NBOH, 34-NBOH e MD-NBOH foram sintetizados. Na metodologia com o reagente colorimétrico cloridrato de 3-metil-2-benzotiazolinona hidrazona (MBTH), foram preparadas soluções acidificadas de MBTH e Fe(NH4)(SO4)2.12H2O. Os resultados foram analisados variando os solventes, concentrações dos analitos e reagentes, bem como os analitos utilizados nos testes. Para a 4-aminoantipirina (4-AAP), foram preparadas três soluções: tampão NH4OH/NH4Cl, 4-AAP e K3Fe(CN)6. Os resultados foram analisados variando as concentrações dos reagentes, ordem de adição, tipos de analitos e tempo de reação empregados nos testes. Após otimização das variáveis estudadas, foi realizada a validação qualitativa do protocolo colorimétrico. Posteriormente, foram realizados estudos eletroquímicos utilizando eletrodo serigrafado de carbono e a técnica de voltametria de pulso diferencial com redissolução adsortiva (AdSVPD) com os analitos e os produtos da reação colorimétrica para confirmação do resultado do teste presuntivo, colocado à prova através da análise de 33 amostras apreendidas fornecidas pela Polícia Civil do Distrito Federal (PC-DF). Para os estudos toxicológicos preliminares, foram realizados estudos com adultos e larvas de peixes Danio rerio, a fim de observar os efeitos de uso crônico por 96h. Durante o desenvolvimento da metodologia colorimétrica, observou-se que o uso de ambos os reagentes colorimétricos possibilitou a diferenciação entre a solução branco e as soluções contendo os analitos, mas apenas a metodologia com 4-AAP foi eficaz na detecção seletiva de NBOHs, onde o produto da reação apresentou uma cor laranja-vermelha, cuja intensidade está diretamente relacionada com a concentração dos analitos, a apresentou limite de detecção de 50 g e alta seletividade, não apresentando resultados falso-positivos quando empregadas amostras que não continham NBOHs. Esse resultado, combinado com a identificação de um pico característico nos voltamogramas obtidos através da técnica de AdSVPD, relativo ao produto cromogênico formado, possibilitou a confirmação dos resultados colorimétricos, além de melhorar significativamente o limite de detecção do protocolo. Durante a análise das fornecidas pela PC-DF, observou-se que o protocolo combinado de análise permitiu a obtenção de quatro perfis eletroquímicos distintos, diferenciando as quatro substâncias geralmente apreendidas na forma de selos de papel (LSD, NBOMes, NBOHs e 2Cs). Em relação aos testes toxicológicos, a substância MD-NBOH produziu letalidade crônica somente na diluição de 100% nos animais adultos, e constataram-se alterações comportamentais semelhantes aos sinais clínicos de intoxicação descritas em seres humanos (agitação, agressividade, medo, perda total e parcial de equilíbrio e convulsão), sobretudo em altas concentrações. No estágio larval foram observadas alterações morfológicas indicativas de toxicidade em todas as diluições avaliadas. Conclui-se, portanto, que a combinação dessas técnicas é muito promissora para a identificação seletiva dessas classes de substâncias em drogas de rua, com foco na obtenção de três respostas analíticas distintas para a presença de NBOHs, facilitando o processo de emissão de laudos periciais, além da necessidade de novos estudos sobre os efeitos fármaco-toxicológicos dessa classe de sustância. |

Abstract |
New Psychoactive Substances (NPS) known as NBOHs are substituted phenethylamines with a structure similar to NBOMes, which makes the development of rapid and selective detection tests for unequivocal identification challenging. Colorimetric tests are advantageous in substance detection due to their low cost, rapid results, use of stable reagents, and potential for in loco application. Furthermore, due to their recent introduction into the illegal market for abuse, few studies describe the potentially toxic effects resulting from the use of these substances. Therefore, this study presents two objectives: 1) to develop a field-application colorimetric method for the selective detection of NBOHs in seized drugs and 2) to conduct initial studies on the toxicity of the substance called MD-NBOH. Initially, the analytes 25H-NBOH, 34-NBOH, and MD-NBOH were synthesized. In the methodology using the colorimetric reagent 3-methyl-2-benzothiazolinone hydrazone hydrochloride (MBTH), acidified solutions of MBTH and Fe(NH4)(SO4)2.12H2O were prepared. Results were analyzed by varying solvents, analyte and reagent concentrations, as well as the analytes used in the tests. For 4-aminoantipyrine (4-AAP), three solutions were prepared: NH4OH/NH4Cl buffer, 4-AAP e K3Fe(CN)6. Results were analyzed by varying reagent concentrations, addition order, types of analytes, and reaction time employed in the tests. After optimizing the studied variables, qualitative validation of the colorimetric protocol was performed. Subsequently, electrochemical studies were conducted using screen-printed carbon electrodes and the Adsorptive Differential Pulse Voltammetry (AdDPV) technique with the analytes and colorimetric reaction products to confirm the presumptive test results, which were tested through the analysis of 33 seized samples provided by the Civil Police of the Federal District (PC-DF). For the preliminary toxicological studies, tests were conducted with adult and larval Danio rerio fish to observe the effects of chronic use over 96 hours. During the development of the colorimetric methodology, it was observed that the use of both colorimetric reagents allowed for differentiation between the blank solution and solutions containing the analytes; however, only the 4-AAP methodology was effective in the selective detection of NBOHs. The reaction product presented an orange-red color, the intensity of which is directly related to the concentration of the analytes, showing a detection limit of 50 g and high selectivity, with no false-positive results when samples without NBOHs were used. This result, combined with the identification of a characteristic peak in the voltammograms obtained through the AdSVPD technique relative to the chromogenic product formed, allowed for the confirmation of the colorimetric results and significantly improved the protocols detection limit. During the analysis of samples provided by the PC-DF, it was observed that the combined analysis protocol allowed for the identification of four distinct electrochemical profiles, differentiating the four substances generally seized in blotter paper form (LSD, NBOMes, NBOHs, and 2Cs). Regarding toxicological tests, the substance MD-NBOH produced chronic lethality only at 100% dilution in adult animals, and behavioral changes similar to clinical signs of intoxication described in humans (agitation, aggressiveness, fear, total and partial loss of balance, and seizure) were observed, especially at high concentrations. In the larval stage, morphological changes indicative of toxicity were observed in all evaluated dilutions. It is concluded, therefore, that the combination of these techniques is very promising for the selective identification of these classes of substances in street drugs, focusing on obtaining three distinct analytical responses for the presence of NBOHs, facilitating the issuance of forensic reports, in addition to the need for further studies on the pharmaco-toxicological effects of this class of substance. |
